Penghargaan ini diberikan untuk menghormati seorang aktor yang telah memberikan penampilan luar biasa dalam peran utama dalam serial televisi musikal atau komedi selama satu tahun kalender.
Penghargaan ini pertama kali diberikan di Penghargaan Golden Globe ke-19 pada tanggal 5 Maret 1962, dengan judul Bintang TV Terbaik – Pria kepada John Charles Daly dan Bob Newhart. Nominasi untuk penghargaan ini diumumkan setiap tahunnya mulai tahun 1963. Penghargaan ini awalnya menghormati aktor dalam genre komedi dan drama hingga tahun 1969, ketika penghargaan tersebut dibagi menjadi beberapa kategori yang menghormati penampilan komedi dan drama secara terpisah. Penghargaan ini dipersembahkan dengan judul baru Aktor TV Terbaik – Musikal atau Komedi dan pada tahun 1980 dengan judul saat ini.
Sejak awal, penghargaan ini telah diberikan kepada 45 aktor. Alan Alda telah memenangkan penghargaan terbanyak dalam kategori ini dengan enam kemenangan dan menerima nominasi terbanyak dengan 11 nominasi.
